| I. Background | |
| 白细胞介素 23 （IL23） 是慢性炎症性疾病（如银屑病、炎症性肠病、多发性硬化症 或类风湿性关节炎）开展的关键促炎细胞因子，与 IL12、IL27、IL35 和 IL39 一起属于 IL12 家族 。过度 IL23 信号转导的病理后果与其促进靶细胞群（主要是分泌 Th17 和 IL17 的 TCRγδ 细胞 （Tγδ17） ）产生炎症介质的能力有关，例如 IL17、IL22、粒细胞-巨噬细 胞集落刺激 （GM-CSF） 或肿瘤坏死因子 （TNFα）。这些致病介质促进粒细胞和巨噬细 胞的募集和活化，导致组织损伤，诱发慢性炎症，最终开展出临床症状。 IL23 是一种异二聚体细胞因子，由两个二硫键连接的结合亚基 p40（由 IL12β编码） 和 p19 （IL23α） 形成 ，它们与 IL12Rβ1 和独特的 IL23R 形成的膜受体复合物结合。IL23 与 IL12 共享 p40 亚基及其受体成分之一 IL12Rβ1。尽管存在相同的亚基，但 IL23 和 IL12 具有不同的生物学功能。IL23 刺激 JAK2 和 TYK2 活性，导致信号转导和转录激活因子 （STAT）的激活。 |
